Not if you do it as multiple smaller regions. I think the size of the area can depend on the PC's hardware & performance. If it's on, say, a netbook, don't even bother. If you have even a decent PC (good processor, probably 4GB RAM) then you should be able to do it in around 100,000 node blocks. I've done 150,000 on my PC.

l can set 40,000,000 nodes with worldedit (with my changes) in less than a second without problems.
